Kv2.2 调控神经系统活动并参与疾病发生发展的研究现状

【摘要】 Kv2.2 是电压门控性钾离子通道 Kv2 家族中的一员，在机体的多个器官中均有表达，是构成神经元膜兴奋性电流的分子基础，能够通过影响动作电位介导神经元的兴奋性并参与早期神经系统的发育。同时，Kv2.2 也参与了神经病理性疼痛、睡眠障碍、肠梗阻、糖尿病等疾病的发生发展，其对神经元内质网/质膜连接的影响以及该通道本身的磷酸化/去磷酸化状态的改变在神经元发挥生理作用中有核心作用。Kv2.2 表达下调时会引起动作电位波形和频率的改变，导致膜兴奋性受到影响从而引起一系列病理生理过程的变化。通过 Kv2.2 干扰病毒干预其在机体内的表达会导致神经元兴奋性的增高，诱导疾病加重，提示 Kv2.2 可能作为疾病治疗的新靶点。本文就近年来关于 Kv2.2 参与疾病发生发展过程进行综述。

【Abstract】 Kv2.2，a member of the Kv2 family of voltage-gated potassium channels，is expressed in many organs of the body and is the molecular basis of neuronal membrane excitatory currents. It can mediate neuronal excitability by affecting action potentials，also participate in the early development of nervous system. At the same time，it is also involved in the occurrence and development of neuropathic pain，sleep disorders，intestinal obstruction，diabetes and other diseases. Its effect on neuronal endoplasmic reticulum/plasma membrane junctions and the change of phosphorylation/dephosphorylation state of the channel itself play a central role in the physiological role of neurons. When the expression of Kv2.2 is down-regulated，the waveform and frequency of action potential will be changed，and the excitability of membrane will be affected，which will lead to a series of pathophysiological changes. Interfering the expression of Kv2.2 with virus will lead to the increase of neuronal excitability and aggravate the diseases，suggesting that Kv2.2 may be a new target for disease treatment. This article reviews the involvement of Kv2.2 in the occurrence and development of diseases in recent years.
